1. Increased energy levels
Amongst its many functions, NAD+ helps your body convert food into energy that it can use. As we grow older, the levels of NAD+ decrease, resulting in lower energy levels in general. Because NMN is a precursor to NAD+, one of the most noticeable benefits of supplementing with NMN is increased energy levels. This can be within a couple of minutes if you take a sublingual NMN that’s absorbed directly into the bloodstream.
2. Improved cardiac function
NMN has been shown to improve vascular function, increase levels of NAD+ in the heart, and also protect the heart from injury and failure. When taking NMN, you will most likely notice improvements in your cardiac functioning and physical endurance. For example, you might be able to run or walk for longer than before or you might find that your heart rate recovers much quicker after cardio exercises. Of course, this will also mean that NMN will help decrease the chances of cardiovascular disease and dysfunction.
3. Regulates blood sugar
NMN also helps regulate blood sugar levels and improves insulin resistance. It may also help regulate cholesterol levels. Taking NMN in conjunction with a healthy diet and active lifestyle will most likely help stabilize your blood sugar levels and decrease your chances of becoming diabetic. When supplementing with NMN, you might well find that you don’t have blood sugar highs and lows throughout the day and that your energy levels remain constant.
4. Weight loss
NAD+ turns on sirtuins which control cell metabolism. Some people have even dubbed SIRTS as being the “skinny gene” because of its metabolism-revving function. Supplementing with NMN was shown to reduce weight gain associated with aging and also helped regulate glucose intolerance. Apart from increased energy, improved cardiac function, and stabilized blood sugar levels, you will most likely also experience weight loss because of your revved up metabolism.
5. Improved brain function
NAD+ is crucial to brain function. The brain uses a tremendous amount of energy and low levels of NAD+ affect the brain’s ability to function optimally. Studies are finding that NMN can benefit brain function and help prevent diseases such as Alzheimer’s by increasing the levels of NAD+ in the brain. Doing so was shown to decrease the build-up of plaques associated with Alzheimer’s, protect brain cells from dying, protect nerve cells from degenerating, and to improve memory.
6. Improved sight
NMN has also been found to improve eye function by preventing the degeneration of photoreceptors and restoring vision. It has also been shown to help with a condition that is related to aging known as dry eye syndrome by increasing tear production. The same researchers also found that NMN improved retinal function. More studies need to be done on this aspect of NMN, but it does show promise. When you supplement with NMN, you may well find that your eyes feel less scratchy and dry, and you might experience improvement in retinal functioning.
7. Slowed aging
One of the most ground-breaking findings of the studies on NMN was its ability to slow down the signs of aging and possibly even reverse them. NMN prevents the expression of genes that control aging and, as mentioned, boosts SIRTS which are very much anti-aging. Interestingly, in one study, it was found that it reversed infertility in aged post-menopausal subjects, making them fertile and able to once more reproduce.
NMN occurs naturally in food such as avocado, broccoli, cabbage, and tomatoes. Now, you might be thinking: “If NMN is found in food, can’t I increase my levels of NAD+ by eating more of those foods? ” That’s a good question and we do encourage you to eat a healthy diet. But, here’s the issue:
Although NMN occurs in these foods, the concentrations are less than 1 mg per kg of food. In other words, to get about 1mg of NMN, you’d have to eat about 1kg of broccoli!
It’s been found that to boost the levels of NAD+ in humans, the levels of NMN should be in the hundreds of milligrams per dose. This is way higher than what we can get from our diets, no matter how much broccoli we eat.

Why Is NMN Expensive?
Pure, high-quality NMN is expensive because of the innovative manufacturing process, as well as all the safety measures that are in place to ensure that the NMN is pharmaceutical grade. To understand the cost behind NMN supplements we need a basic understanding of the manufacturing process.
NMN is made through enzymatic synthesis. The nicotinamide molecule goes through 6 complete stages before the NMN molecule is produced. After NMN is finally produced there are a further 7 stages before completing the final product. These including purification, concentration, crystallization, filtering, drying and grinding, testing with quality control, and the packaging of the final product.
To simplify the manufacturing process there are two stages:
Nicotinamide → nicotinamide riboside (NR), and then nicotinamide riboside → β-nicotinamide mononucleotide (NMN).
Let’s take a closer look:
● Nicotinamide → NR: After adding ribose and water to nicotinamide, and then adding nicotinamide phosphoribosyltransferase, NR is finally obtained through the catalytic reaction of nicotinamide phosphoribosyltransferase.
● NR → NMN: We then add adenosine triphosphate and nicotinamide riboside kinase to NR after a nicotinamide riboside kinase catalytic reaction. This is followed by hydrolyzation → extraction → purification → concentration → recrystallization → drying and grinding → quality assurance and checks → NMN.
Reverses oxidation and improves age-related blood flow issues
The process of aging and many of its age-related complications are directly linked to the depletion of NAD+ in our bodies. The restoring of NAD+ levels have shown to increase the lifespan of subjects. NMN being the precursor of NAD+, can provide these beneficial effects as well. Deterioration of the vascular system and oxidative stress are common occurrences during the aging process. NMN proved to effectively reverse vascular depletion and oxidative stress.
Decreases age-associated weight gain
Age-associated increase in body weight and obesity-related complications, including a decrease in energy metabolism and locomotor activity, age-dependent insulin insensitivity, and higher triglyceride levels were reversed by a 12-month NMN intervention. NMN’s success in this preclinical study prompted researchers of Washington University School of Medicine of St. Louis and Keio University School of Medicine in Tokyo to commence a collective research program for the phase I NMN clinical trial. A positive outcome of this study could curve a new direction for anti-aging treatment strategies.
May help treat diabetes
NMN has shown its potential as a treatment for type 2 diabetes. The primary characteristic of type 2 diabetes is insulin resistance which is caused by oxidative stress and various other factors. These are all corrected by NAD+. A fatty diet and aging contributes greatly to this type of diabetes and is usually linked to the reduction of NAD+. To correct thehigh-fat diet and aging-induced type 2 diabetes NMN was administered in high-fat, diet-fed female and male diabetic subjects. All of them showed significant improvements in insulin intolerance. In the case of age-induced diabetes, NMN corrected glucose intolerance in both male and female diabetic mice.
May help with Alzheimer’s Disease and Intracerebral Hemorrhaging
NMN has shown promise in the treatment of Alzheimer’s as it focuses on and nurses the causes of this disease. A recent study demonstrated its ability to directly target and treat the underlying causes namely a decrease inoxygen consumption rates (OCR) and NAD+. Intracerebral hemorrhage (ICH), bleeding on the brain, is a neurological disorder responsible for 10-15% of all strokes. NMN may improve the condition. A study oncollagenase-induced ICH showed that NMN treatment increased intracerebral NAD+ concentration post-ICH and provided protection against amyotrophic lateral sclerosis (ALS or motor neurone disease) and ischemic stroke.
NMN may help prevent ischemia-reperfusion injury
Ischemia occurs when a part of your body does not get sufficient blood. Ischemic events cause the amount of oxygen and adenosine triphosphate (ATP) levels found in cardiac muscles to decrease. Reperfusion is the process of resupplying blood to these tissues which have gone through ischemia. Ischemia followed by reperfusion is a deadly condition dealing with severe tissue damage. Ischemic preconditioning or IPC is the body's way of counteracting this deadly condition.

Just like finding the correct dosage, there is conflicting evidence surrounding the time of day to consume NMN. It seems age may also play a part in deciding when to take NMN.
Research completed by the University of Waterloo in Canada involving a model that simulates the circadian clock and metabolism based on the mouse liver suggests that the best time younger adults should consume NMN is six hours after they wake up.
This differs from a study completed by the University of Tsukuba in Japan involving healthy individuals over the age of 65 that were split into four random groups. One group received 250 mg NMN in the morning before noon, the second group 250 mg NMN in the evening following 6 p.m., plus two correlating placebo groups.
The study identified four areas, including characteristics of participants, sleep quality, fatigue, and physical performance throughout 12 weeks. Significant discoveries were found only in the fatigue and physical performance results, with a group taking NMN in the evening, which showcased superior results over tested groups.
The studies mentioned regarding NMN timing and dosage can not be generalized for all populations. These studies focus primarily on results from Asia, older men, athletes, women with prediabetes, and simulated models based on mice liver analysis. More large scale studies are needed to understand the exact benefits of NMN.
While no side effects have been reported, there is still no consensus on the right amount of NMN. Many studies to date have looked at low doses. The 2022 clinical study assessing results of placebo, 300, 600, and 900mg doses found the greatest efficacy in the 900mg group.
The endurance trial investigating doses of 300, 600, and 1,200mg daily found the biggest impact in the medium and high groups, which averages a 900mg daily dose.
Supplement providers will often recommend correct levels based on their research and observations. Some will suggest varying the doses by weight with higher doses recommended for heavier people.
NMN supplements will often come in sizes between 100mg and 500mg with doses of between one and six tablets recommended. Check the manufacturer’s label to see how much they recommend taking.
Clinical understanding of NMN supplements is still evolving, with each trial and piece of research enhancing our knowledge further. From what has been observed in the studies conducted, it seems that this approach provides actual advantages in relation to health and lifespan, with minimal occurrences of side effects.
Is NMN an Anti-Aging Molecule?
While many stories in the media characterize nicotinamide mononucleotide as an anti-aging molecule, no human studies support this idea. Right now we know that NMN is safe and boosts NAD+ levels. Animal and human studies demonstrate that NMN supplementation offers benefits related to oxygen utilization, athletic endurance, and muscle insulin sensitivity. The science of NMN supplementation is evolving fast, however, as researchers seek to understand other ways that restoring levels of the essential molecule NAD+ can support healthy aging. Meanwhile, several other clinical trials are underway, studying fatigue associated with COVID-19, cardiometabolic function, organ system biology, and more.
How Does NMN Work?
NMN, or nicotinamide mononucleotide, is a cellular molecule that’s considered a precursor to NAD+. This means NMN will chemically transform into NAD+ in your cells. When you think about NMN vs. NAD+, remember that these two molecules work together to metabolize energy in your cells.
After NMN is absorbed into your bloodstream, it converts into NAD+. Then the NAD+ is stored in your muscles and vital organs, helping them function in peak condition.
One of the fastest ways to stimulate your body’s NAD+ production is to increase levels of NMN. There is evidence that NMN’s boost to NAD levels may be able to improve health in the heart, liver, muscles, and kidneys, and even slow down the progression of diabetes and Alzheimer’s.
Beyond their role in making NAD+, NMN molecules have their own health benefits. NMN supplements can aid your body’s insulin production and glucose tolerance. They have positive effects on overall metabolism as well.
NMN supplements are the easiest way to raise NMN levels. Low volumes of NMN are also found in certain foods like broccoli, avocado, cucumber, and cabbage.
One of the most notable benefits of NMN is that it facilitates DNA repair through the activation of sirtuins, a family of proteins that regulate cellular health, metabolism, and DNA repair.
Specifically, SIRT1, one of the most studied sirtuins, plays a crucial role in the repair of damaged DNA. SIRT1 relies on NAD+ to function effectively; when NAD+ levels are sufficient, SIRT1 can activate and promote the expression of genes involved in DNA repair and cellular maintenance.
This activation is essential for managing oxidative stress and repairing DNA breaks, which can occur due to environmental factors, metabolic processes, or normal cellular aging.
Furthermore, NMN’s role in enhancing the activity of poly (ADP-ribose) polymerase 1 (PARP1) is also significant for DNA repair.
PARP1 is an enzyme involved in detecting and repairing single-strand breaks in DNA. When DNA damage occurs, PARP1 uses NAD+ to form poly(ADP-ribose) chains that recruit other repair proteins to the site of damage, initiating the repair process.
By increasing NAD+ levels, NMN enhances the activity of PARP1, thereby improving the cell’s capacity to respond to DNA damage effectively.
This is particularly important for maintaining genomic integrity and preventing the accumulation of mutations that can lead to age-related diseases, leading to the possibility that NMN may reduce or reverse ageing.
Bioavailability and Absorption
While both NMN and NR effectively increase NAD+ levels, they differ in how the body absorbs and utilizes them.
NMN is a larger molecule and may be absorbed differently in the gastrointestinal tract.
Some studies suggest that NMN may be more efficiently taken up by tissues, potentially leading to faster increases in NAD+ levels.
On the other hand, NR is generally well-absorbed and can effectively raise NAD+ levels, but may require more metabolic steps before converting into NAD+.
Specific Health Goals
For individuals primarily interested in enhancing energy metabolism and physical performance, both NMN and NR can be beneficial, but it is more recommended to buy NMN as it has more of an edge due to its potential for quicker absorption and direct conversion to NAD+.
Those seeking to improve metabolic health, support weight management, or increase exercise capacity might find NMN more effective.
If cognitive benefits are the primary goal, both NMN and NR can enhance brain function by increasing NAD+ levels, but individual responses can vary.
Some studies suggest that NMN may provide additional neuroprotective benefits due to its rapid action in the brain, while NR also demonstrates cognitive enhancement capabilities.
Age and Health Status
Older adults or individuals experiencing age-related declines in NAD+ levels might benefit more from NMN supplementation.
Research has shown NMN’s effectiveness in improving muscle function, enhancing physical activity, and supporting overall vitality in aged models.
For younger individuals, particularly those who are active or looking to support general wellness, both NMN and NR are effective.
Lifestyle Considerations
Individuals with specific dietary restrictions or sensitivities may also factor into the choice between NMN and NR.
For instance, NMN is typically found in lower concentrations in foods compared to NR, which is present in dairy products and some fermented foods.
This may lead some people to prefer NMN for supplementation, particularly if they follow a vegan or lactose-free diet.
